Spesies Sulfolobus tumbuh di mata air vulkanik dengan pertumbuhan optimal terjadi pada pH 2-3 dan suhu 75-80°C, membuat mereka asidofili dan termofili masing-masing. Sel-sel Sulfolobus berbentuk tidak teratur dan flagelar.
