Amal Alamuddin Clooney lahir di Beirut, Lebanon. Nama depannya berasal dari أمل, ʾyang berarti "harapan" dalam bahasa Arab. Ayah Amal, Ramzi Alamuddin, adalah seorang pensiunan akademisi, dan ibunya, Baria, merupakan jurnalis.[10] Keluarganya meninggalkan Lebanon karena perang saudara saat Amal masih berusia tiga tahun.[11] Mereka kemudian menetap di Gerrards Cross, Buckinghamshire. Amal mempunyai tiga saudara kandung: satu saudara perempuan (Tala Alamuddin) dan dua saudara tiri dari pernikahan pertama ayahnya.[12]
